Ensaios com vista à indução de embriogénese somática em oliveira (Olea europaea L.)

Abstract

O trabalho experimental realizado no âmbito desta dissertação teve como objetivo definir um protocolo para indução e manutenção de embriogénese somática em oliveira (Olea europaea L. ssp. europaea var. sativa). Para isso, utilizou-se como material inicial; - pecíolos de folhas provenientes de material em cultura in vitro; - estames e carpelos recolhidos em inflorescências de árvores adultas; - radículas e cotilédones provenientes de sementes maduras. Em qualquer destes casos foi utilizada a cultivar a ‘Galega vulgar’, sendo que, no caso dos pecíolos, o material é de natureza clonal, cl. 1441, e, no que às sementes diz respeito, foram recolhidas em árvores adultas de ‘Galega vulgar’ em polinização livre. Em todos os ensaios, a definição das condições de cultura, assim como dos meios nutritivos, teve em conta o trabalho realizado com sucesso por outros autores, nesta mesma espécie, inclusive nos ensaios com estames e carpelos, onde até agora não há referência a resultados positivos em oliveira. Os protocolos propostos por estes autores foram testados e os resultados obtidos avaliados e comparados com o trabalho original. Em alguns casos procedeu-se ainda a um trabalho exploratório, com alteração das variáveis em estudo, ou dos seus níveis, na expetativa de melhorar os resultados obtidos. Destes, salienta-se o seguinte; - indução de calogénese em todos os tipos de explantes utilizados; - obtenção de embriogénese somática, de uma significativa taxa de conversão dos embriões em plantas e de embriogénese repetitiva, quando o material juvenil foi utilizado; - obtenção de rebentos neoformados sobre calli de pecíolos. A obtenção de bons resultados com os materiais de natureza juvenil, assim como a obtenção de organogénese sobre calli em materiais adultos, deixa antever possibilidade de definição de um protocolo eficiente de embriogénese somática nesta cultivar, com a aplicação de ligeiras adaptações aos protocolos agora testados; ABSTRACT: The experiments carried out for this dissertation aimed on establish a protocol for induction and maintenance of somatic embryogenesis in Olive (Olea europaea L. ssp. europaea var. sativa). To achieve this goal, the following start materials were used; - leaf petioles from in vitro cultured explants; - stamens and carpels collected from inflorescences of adult trees; -rootlets and cotyledons from ripe seeds. The cultivar ‘Galega vulgar’ was used in all trials. The petioles have a clonal origin, ‘Galega vulgar’ cl. 1441, and, the seeds and flowers were collected from mature trees in open pollination orchards. The culture media, as well as the culture conditions here used, always considered the successfully work reported by other authors, within the same species, including the essay with stamens and ovaries, where, to the best of our knowledge, there are no reference to positive results in Olive. The protocols proposed by these authors were tested and the achieved results evaluated and compared with the original work. In some cases, exploratory trials aiming on improve the results were also carried out, both, by changing the variables under evaluation or their levels. After data analyses, the following results may be highlighted; - callogenesis induction on all kinds of explants tested; - somatic embryogenesis, good embryo to plant conversion, and repetitive embryogenesis, when the juvenile materials were used; - shoot organogenesis over calli from petioles. The good results achieved with the materials of juvenile nature, as well as the induction of organogenesis over calli in adult materials, allow us to believe that the definition of an efficient protocol for somatic embryogenesis using mature explants of this cultivar, relies on slight adaptations to the protocols now tested.
