Abstract: | Resumo - Este trabalho pretendeu avaliar a biodiversidade do agente causal da antracnose (Colletotrichum spp.) no cajueiro (Anacardium occidentale L.), cultivado no Brasil, de modo a contribuir para o delineamento de estratégias de optimização do controlo da doença, quer a nível de resistência da planta hospedeira, quer a nível epidemiológico no campo. Para atingir aquele objectivo recorreu-se a testes de patogenicidade para avaliar a interacção patógeno- hospedeiro, e a análises isoenzimáticas, serológicas e moleculares para estudo de 18 isolados de Colletotrichum spp. obtidos de cajueiro. No estudo da interacção patogénio-hospedeiro, os testes de patogenicidade diferenciaram os isolados por índices de agressividade, em relação à maior ou menor susceptibilidade dos clones de cajueiro utilizados. As inoculações cruzadas entre isolados de Colletotrichum, provenientes de cajueiro e inoculados em frutos de oliveira, e isolados de Colletotrichum provenientes de oliveira e inoculados em folhas de cajueiro, revelaram não haver especificidade destes isolados relativamente aos hospedeiros utilizados (cajueiro e oliveira). Todos os isolados se mostraram patogénicos. A análise isoenzimática permitiu verificar a diversidade existente entre os isolados, relativamente a dez enzimas (esterase-EST, fosfatase ácida-ACP, fosfatase alcalina-AKP, fosfoglucomutase-PGM, fosfoglucoisomerase-PGI, 6-fosfogluconato desidrogenase-PGDH, hexoquinase-HK, isocitrato desidrogenase-IDH, malato desidrogenase-MDH e superóxido dismutase-SOD), tendo-se identificado dois grandes grupos, um constituído maioritariamente por C. gloeosporioides e outro por C. acutatum. A análise serológica dos isolados de Colletotrichum spp., pela técnica PTA –ELISA (Plate trapped antigen-enzyme linked immunosorbent assay) aplicando anticorpos monoclonais específicos para C. acutatum, permitiu identificar, pela primeira vez, isolados de C. acutatum obtidos de tecidos do cajueiro com sintomas de antracnose. A caracterização molecular dos isolados de C. gloeosporioides e C. acutatum, utilizando iniciadores específicos, veio confirmar e clarificar os resultados obtidos pelos testes acima descritos, permitindo uma identificação interespecífica dentro da colecção de isolados de Colletotrichum submetida a análise neste trabalho.
### Abstract - This work was to evaluate the biodiversity of the causal agent of the anthracnose (Colletotrichum spp.) in the cashew tree (Anacardium occidentale L.), farmed in Brazil, so as to contribute for the strategy delineation of the optimisation of control of the disease, whether at the resistance level of the host-plant, whether at epidemiologic level in the field. In order to achieve that objective, we used pathogenecity to evaluate the host-pathogen interaction and isoenzimatic, serological and molecular analysis to study 18 isolates of the Colletotrichum sp. obtained from the cashew tree. In the host-pathogenic interaction study, the pathogenecity tests differentiated the isolates by its aggressiveness in relation to susceptibility of the cashew tree clones used. The crossed-inoculations between the Colletotrichum isolates, from the cashew tree and inoculated in the olive fruits and Colletotrichum isolates from the olive tree and inoculated in cashew leaves, revealed non-specificity of these isolates in relation to the, hosts used (cashew and olive). All the isolates showed to be pathogenic. The isoenzimatic analysis enabled to verify the existing diversity among the isolates, in relation to ten enzymes (esterase-EST, acid phosphatase-ACP, alkaline phosphatase-AKP, phosphoglucomutase-PGM, phosphoglucoisomerase-PGI, 6-phosphogluconate dehidrogenase-PGDH, hexokinase-HK, isocitrate dehidrogenase-IDH, malate dehidrogenase-MDH and superoxide dismutase-SOD), two big groups were identified, one mainly made up by C. gloeosporioides and another mainly by C. acutatum. The serological analysis of the isolates of the Colletotrichum sp., through the PTA – ELISA technique (Mate trapped antigen-enzyme linked immunosorbent assay) applied specific monoclonal antibodies for C. acutatum, identified, for the first time C. acutatum isolates from cashew tree tissues with anthracnose, symptoms. The molecular characterization of the C. gloeosporioides and C. acutatum isolates, using specific primers, confirmed and clarified the results obtained through the above mentioned tests, allowing an inter-specific identification within the Colletotrichum isolates collection submitted to analysis in this work. |
