Comparação de antigénios de Leishmania infantum em testes serológicos

Abstract

Esta dissertação teve como objectivo o desenvolvimento e padronização de um ELISA para diagnóstico da LVC. O estudo foi desenvolvido de acordo com 2 objectivos cujo desenho experimental, resultados e discussões se encontram expostos nos capítulos 2 e 3. No capítulo 1 apresenta-se uma introdução teórica sobre a LVC, com incidência na resposta imunológica e nos métodos de diagnóstico. No capítulo 2, avaliamos o desempenho de dois antigénios (β-ME-DAT e TRYP-DAT), no DAT e conclui-se que o β-ME-DAT tem maior sensibilidade e a mesma especificidade. Assim, o β-ME–DAT foi utilizado como teste referência para o desenvolvimento do trabalho apresentado no capítulo seguinte. O capítulo 3 relata o desenvolvimento do β-ME-ELISA e comparamo-lo com o convencional TRYP-ELISA. O β-ME-ELISA dispõe de maior sensibilidade e especificidade e constitui um promissor método diagnóstico da LVC, sobretudo pelo seu potencial em detectar infecções precoces, e poderá vir a ser utilizado em inquéritos epidemiológicos e/ou rotina clínica; ### ABSTRACT: Comparison of antigens of Leishmania infantum in serological tests The objective of this thesis was to develop and standardize an ELISA able to diagnose CVL. The study was carried out according to two objectives and its experimental design, results and discussions are described in chapters 2 and 3. In chapter 1 we present a theoretical introduction dealing with CVL with relevance to the immune response and specific diagnostic methods. In chapter 2 we evaluated the performance of two antigens (β-ME-DAT and TRYP-DAT) in the DAT and we concluded that the β-ME-DAT has higher sensitivity and same specificity. Accordingly, the β-ME-DAT was used as reference test in the following studies. In chapter 3 we present the development of the β-ME-ELISA and we compare its performance with the one of the traditional TRYP-ELISA. β-ME-ELISA showed higher levels of sensitivity and specificity, reason why, in our opinion it constitutes an encouraging diagnostic CVL tool, especially due to its eventual potential to detect early infections, before appearance of overt disease and subsequent importance in epidemiological studies as well as in clinic routine services.
