Title: New Insights into Somatic Embryogenesis Efficiency in Olea europaea L
Authors: Pires, Rita Nobre
No entanto, a
recalcitrante; Somatic embryogenesis (SE) has emerged as an alternative strategy for the clonal propagation of olive
trees, particularly for genotypes characterised by high recalcitrance to adventitious root development
when propagated using conventional methods. However, the high recalcitrance of adult tissues to the
induction of embryogenic structures continues to limit the application of this technology. Therefore,
understanding the molecular mechanisms associated with the embryogenic response is of particular
importance for the development of alternative propagation systems applicable to the national olive
industry. SE is a morphogenic response induced by exogenous stimuli that involves cellular
reprogramming at the molecular level, reflected in biochemical and physiological changes that
culminate in the differentiation of embryogenic structures. During this process, the interaction between
explants and the culture medium is crucial, including exposure to biomolecules secreted by plant tissues.
This exchange of molecules, particularly proteins and metabolites, between intracellular and
extracellular compartments may contribute to the modulation of the embryogenic response. The present
study aimed to identify mechanisms involved in SE efficiency in olive through the characterisation of
the proteome of high-efficiency (HE) and low-efficiency (LE) embryogenic lines and their
corresponding secretome by LC–MS/MS, complemented by exometabolome analysis using ¹H NMR.
The ability of secreted biomolecules to induce the embryogenic response was assessed by applying
conditioned medium (CM) from HE lines to LE lines and adult material. The results revealed a higher
abundance of proteins associated with vesicular transport, signalling, and cellular reorganisation in HE
lines, which also exhibited, at the extracellular level, proteins related to redox regulation and cell wall
dynamics, together with more efficient carbon utilisation. Application of the CM validated the role of
secreted molecules in the embryogenic response by promoting increased cell proliferation and the
formation of embryogenic structures in recalcitrant material.

Estes resultados indicam a necessidade de estratégias de prevenção específicas, como a segurança alimentar, a biossegurança e a comunicação dos riscos, salientando a importância da abordagem “Uma Saúde” para a vigilância e o controlo da infeção por HEV nos meios rurais mediterrânicos; Hepatitis E in the Alentejo Cork Oak Forest – A study from an integrated "One Health" perspective - Abstract: Hepatitis E virus (HEV) is an emerging zoonotic agent that has been gaining importance in agro-silvo-pastoral ecosystems such as the Montado, where there is a regular interaction between domestic animals, wild animals and humans. The aim of this study was to determine the presence of HEV in animal and environmental reservoirs, as well as to identify potential risk factors for its transmission. Molecular detection by RT-qPCR allowed the analysis of 335 samples of liver (hunted and road killed animals), faeces from wild and domestic animals, and water. The HEV genome was identified in 29 hepatocellular samples, mainly from wild boar, with a positivity rate of 12.22%, followed by fallow deer (7.40%) and red deer (6.97%). Pooled water or faecal samples did not yield positive results. The results confirm that wild boar is the main reservoir, with a heterogeneous spatial distribution. These results indicate the need for specific prevention strategies, such as food safety, biosecurity and risk communication, highlighting the importance of a ‘One Health’ approach for the surveillance and control of HEV infection in rural Mediterranean areas.

Os resultados destacam a necessidade de intervenções integradas One Health para reduzir a transmissão; Taeniasis/cysticercosis: an epidemiological study of knowledge and practices among the population of cape verde - Abstract: Taenia solium tapeworm infection and cysticercosis remain a significant zoonoses, but data from Cabo Verde are limited. This study assessed Knowledge, Attitudes and Practices (KAP) regarding T. solium in rural and peri-urban areas through a cross-sectional survey. Data were collected using structured questionnaires and analysed using descriptive statistics and content analysis. Participants demonstrated general knowledge about the parasite, particularly regarding cysts in pigs, but this knowledge proved to be fragmented, with gaps regarding transmission, routes of infection and clinical manifestations. Although attitudes towards food safety were generally appropriate, unsafe practices persisted, such as informal slaughter and low recourse to veterinary inspection. Understanding of transmission was based on observable environmental factors, indicating limited understanding of the parasite’s life cycle. The results highlight the need for integrated one health interventions to reduce transmission.
